Summary Salmonella enterica subspecies (Salmonella ssp.) are the second most common foodborne pathogens. Cleaning and in particular disinfection is crucial to preventing the spreading of this pathogen in the food processing plants, and therefore it would be devastating for food safety if the bacteria would become resistant to the disinfectants used. Another aspect is the concern that exposure to disinfectants might lead to decreased susceptibility to antibiotics or risk of crossresistance between biocides and antibiotics. The aim of this PhD project was to determine the prevalence of biocide resistance in Salmonella ssp. and to elucidate mechanisms of resistance. The susceptibility of Salmonella ssp. isolated from Danish slaughterhouses to two disinfectants (Incimaxx DES containing mainly oxidizing agents and Desinfect Maxi containing quaternary ammonium chloride compounds (QACs)) and triclosan a widely used antibacterial agent, was examined together with antibiotic susceptibility. No biocide resistant strains were found, but comparing MICs of isolates obtained before cleaning with those obtained after indicated that cleaning and disinfection might select isolates with higher MICs to Incimaxx DES. No correlations were found between time of sampling and antibiotic resistance and only weak correlations, if any, were found between minimum inhibitory concentrations (MICs) to biocides and MICs to antibiotics. Preliminary results from examinations of growth curves of 20 strains of Salmonella enterica ssp. enterica serovar Enteritidis (S. Enteritidis) and S. Typhimurium when exposed to concentrations of biocides below MICs suggest concentration-dependent increase in lag-time and decrease in specific growth rate. Regrowth, if any, after biocide exposure was not affected. Decreased susceptibility to triclosan was observed after continuous exposure to triclosan in S. Typhimurium and E. coli. Adaptation of S. Typhimurium and E. coli by continuous exposure to Desinfect Maxi and Incimaxx DES was not possible with the method used. Cross resistance between triclosan and enrofloxacin together with triclosan and sulfamethoxazole/trimethoprim were observed for the triclosan adapted strains of S. Typhimurium when they were concurrently exposed to triclosan. The mechanism responsible was suggested to be increased efflux pump activity.
Resistance mechanisms of the triclosan adapted strains were examined and revealed that mutations in fabi in combination with other mutations, e.g. mutation in rpos or rpod and/or ndh confer high level resistance to triclosan. In conclusion, the prevalence of biocide resistant Salmonella on Danish slaughterhouses is low and continuous exposure to disinfectants does not seem to induce resistance. However, continuous exposure to triclosan can induce resistance to triclosan and concurrent exposure of these strains to high concentrations of triclosan can confer cross-resistance to some antibiotics probably due to increase in efflux pump activity. Further examinations are necessary to elucidate the precise mechanisms of resistance, but the evidence of cross resistance between biocides and antibiotics due to common mechanisms of resistance underlines the necessity of continuous monitoring.
Sammendrag Salmonella enterica subspecies (Salmonella ssp.) er en væsentlig årsag til fødevarebåren sygdom. Rengøring og især desinfektion er vigtig for at undgå spredning af dette patogen på fødevarevirksomheder. Derfor ville det også være katastrofalt for fødevaresikkerheden, hvis bakterier blev resistente over for de desinfektionsmidler, der bliver brugt. Derudover er der bekymring for, at der kan være kryds-resistens mellem desinfektionsmidler og antibiotika, eller at bakterierne efter kontakt med desinfektionsmidler kan blive mindre modtagelige over for antibiotika. Formålene med dette Ph.d. projekt var at undersøge prævalensen af biocid-resistens i Salmonella ssp. samt at beskrive mekanismerne for resistens. Salmonella ssp. blev isoleret fra danske slagterier og deres følsomhed overfor to desinfektionsmidler (Incimaxx DES, som indeholder oxiderende stoffer og Desinfect Maxi som indeholder kvarternære ammonium forbindelser (QAC)) samt triclosan, et antibakterielt stof, som bliver brugt i mange forskellige produkter, og antibiotika blev undersøgt. Der blev ikke fundet nogen isolater, som var resistente overfor desinfektionsmidlerne eller triclosan, men når man sammenlignede MIC-værdierne for isolater indsamlet før rengøring med dem indsamlet efter, var der indikationer på at rengøring og desinfektion kan selektere for isolater med nedsat følsomhed over for Incimaxx DES. Der blev ikke fundet nogen korrelationer mellem antibiotika resistens og om isolatet var indsamlet før eller efter rengøring, og der blev kun fundet enkelte, svage korrelationer mellem MIC overfor biociderne sammenlignet med MIC overfor antibiotika. Biocidernes indvirkning på vækst blev undersøgt på 20 stammer af Salmonella enterica ssp. enterica serovar Typhimurium (S. Typhimurium) og S. Enteritidis. Foreløbige resultater tyder på, at lave koncentrationer af biociderne, medfører en koncentrationsafhængig forlængelse af lag-fasen og nedsættelse af vækstraten, men at væksten uden biocider efterfølgende ikke var påvirket. Der blev observeret krydsresistens mellem triclosan og sulfamethoxazol/trimethoprim samt triclosan og enrofloxacin i S. Typhimurium stammer der var adapteret til triclosan, men kun når stammerne samtidig blev eksponeret til høje koncentrationer af triclosan. Mekanismen bag blev foreslået at være øget aktivitet af efflux pumper.
Adaptation af S. Typhimurium og E. coli ved kontinuerligt at lade dem vokse med stigende koncentrationer af Desinfect Maxi og Incimaxx DES førte ikke til nedsat følsomhed over for disse desinfektionsmidler. Mekanismerne for triclosan resistens blev undersøgt, og viste at mutationer i fabi i kombination med mutationer i andre gener, f. eks. rpos eller rpod og/eller ndh medfører resistens mod høje koncentrationer af triclosan. Afslutningsvist: prævalensen af biocid resistens i Salmonella ssp. fra danske svineslagterier er lav og kontinuerlig eksponering for desinfektionsmidler ser ikke ud til at kunne inducere resistens. Dog kan kontinuerlig vækst med triclosan medføre resistens overfor triclosan, og hvis de adapterede stammer udsættes for antibiotika og høje koncentrationer af triclosan samtidig, er de resistente overfor nogle antibiotika. Dette skyldes formentligt øget pumpe-aktivitet ud af cellen, og derfor er dette et eksempel på kryds-resistens som følge af ens resistens-mekanismer. Yderligere undersøgelser er nødvendig for at klarlægge de præcise mekanismer for resistens over for triclosan og indikationerne på kryds-resistens tydeliggør nødvendigheden for fortsat overvågning.
